100 mM Ribonucleoside Triphosphates (NTPs)

1. Dissolve each NTP in DEPC-treated H2O at a concentration of 200 mM.

2. Dilute an aliquot of the NTP appropriately, and read the optical density at the wavelengths given in the table below.

3. Calculate the actual concentration of each NTP.
  • For a cuvette with a path length of 1 cm, Absorbance = e/M.

    4. Dilute the solutions with H2O to a final concentration of 100 mM NTP.

    5. Store each separately at -70oC in small aliquots.
