Staining Paraffin Sections with Toluidine Blue

INTRODUCTION

The main advantage of staining using toluidine blue is the short time, required for processing. The sectioned material can be stained without deparaffinization, thus eliminating the hydration-dehydration steps.




MATERIALS AND SOLUTIONS

Toluidine Blue Staining Solution (10 ml)
0.05% Toluidine Blue O ---------------------------- 0.5 ml of 1% Toluidine Blue
Distilled H2O --------------------------------------- 9.5 ml
{or, 0.1 M Citrate Phosphate Buffer (pH4-6) ------- 9.5 ml}




PROCEDURES

1. Stain Non-deparaffinized slides in Toluidine Blue Staining Solution for 15-30 min.

2. Rinse slides in distilled water briefly and air-dry.

3. Remove paraffin with two changes of xylene overnight.

4. Mount slides with synthetic resin
