The main advantage of staining using toluidine blue is the short
time, required for processing. The sectioned material can be stained
without deparaffinization, thus eliminating the hydration-dehydration
steps.
MATERIALS AND SOLUTIONS
Toluidine Blue Staining Solution (10 ml)
0.05% Toluidine Blue O ---------------------------- 0.5 ml of
1% Toluidine Blue
Distilled H2O --------------------------------------- 9.5 ml
{or, 0.1 M Citrate Phosphate Buffer (pH4-6) ------- 9.5 ml}
PROCEDURES
1. Stain Non-deparaffinized slides in Toluidine Blue Staining
Solution for 15-30 min.
2. Rinse slides in distilled water briefly and air-dry.
3. Remove paraffin with two changes of xylene overnight.
4. Mount slides with synthetic resin
