Deionization of Formamide, Formaldehyde, and Glyoxal
by AG 501-X8 Mixed Bed Resin

INTRODUCTION

The rapid oxidation of formamide, formaldehyde, and glyoxal requires deionization prior to use for optimal efficacy in hybridization and nucleic acid denaturation applications. AG 501-X8 resin is a 1:1 equivalent mixture of AG 1-X8 resin (OH-) and AG 50W-X8 resin (H+).




MATERIALS AND SOLUTIONS

AG 501-X8 mixed bed resin (Bio-Rad).




PROCEDURES

1. Add 1 g of AG 501 -X8 resin to each gram of material to be deionized.

2. Mix the solution by magnetic stirrer for 1 hour.

3. Filter twice through Whatman No. 1 filter paper.

4. Dispense into 1 ml aliquotes and store at -20oC.


  • Once opened, the unused portion of a previously deionized aliquot should be discarded.
